When do I pick this fruit, open it up and dry seeds?

I suspect when it falls off (or you can very easily pull it off, but don't force it), or when it cracks open. Whatever comes first.

As Aiko says.

Are you sure this is fertilised? Cereus fruits tend to be fairly large, plump and round.

Usually the flowers will immediately dry up and fall off. Then the lower portion of the flower would dry up and fall off a few days later. This the first time the lower portion stayed and has gone from green to this deep purple.
Here it is right after it bloomed. It has grown considerably in size as it has matured.

Corrected. A day after it bloomed.
